Theory question here:
I thought a ROM was basically the operating system, totally replacing whatever the manufacturer default is. Why then are certain ROMs only good for certain Android versions?
because some use framework from that version... and some... Because of the locked bootloader... Run off that kernel... And different manufacture versions have different kernels

Will all of these work if I am coming from .621? I am currently running Vortex but want to try Tranquility.

Nope all of the ones listed here except for the first one are Froyo, you would need to SBF to Froyo to flash, can't do that once on .621 (the no reverting to earlier versions thing).
The very first one may work but you would need the .zip patched otherwise you'll get a panic and bootloader error out. (And there's really no reason to want that as its a bit outdated at this time anywho)
Are there any good roms out that will work on the stock .621 rooted via VooDoo OTA? I have tried Vortex, but just un-stable, and I'm not a big Miui fan.....
Is it true that any 2-INIT rom will run on the stock rooted .621?

not a miui fan? in my opinion its one fo the best stable roms out there im using beanstown now, waiting for a wizard release hopefully soon if if decides to do another big fan of the Wiz, i use boot manager with vortex, liberty, liquid, cm9 and steel i always go back to miui though, cm9 works good but im not sure about .621 in never did the ota. vortex rc1 is good there are good roms out there just look try rootz wiki
