Only if the app requires a data connection. The only time you use data is when you surf the web, download from the web, use apps to download like Pandora or ultimate mp3. Why do you ask anyway?

If you have a limit on the data you use then download firewall from the market.I think its free and it allows only needed info to pass through.

When you're on Wifi, it doesn't use your data, it uses the wifi for data. It likely was something using data while not on Wifi. Maps and email use data to function, so maybe it was something like that while on 3g.
Do you not have unlimited data?

Download TrafficStats lite from the appstore. It will show all data use, both sent and received, by all apps on a per app basis. Remember, a lot of free apps show advertising which it gets from the net, and will show data usage for that app
